摘要 目的检测维生素E琥珀酸酯(VES)对MCF-7乳腺癌细胞三苯氧胺敏感性的影响。方法人乳腺癌细胞MCF-7以VES刺激24h,VES浓度为5μg/ml和10μg/ml。随后以1μM,2.5μM,5μM和10μM的三苯氧胺作用24h,以MTT法测定对细胞增殖的抑制作用,RT-PCR法检测VES对HER-2mRNA表达的影响,以免疫组化法检测VES作用前后HER-2产物c-erbB-2表达的变化。结果TAM对MCF-7乳腺癌细胞具有增殖抑制作用,VES处理后,乳腺癌细胞对TAM的敏感性升高。RT-PCR显示VES作用后乳腺癌细胞HER-2mRNA转录水平有一定程度的下降,乳腺癌细胞c-erbB-2的表达降低。结论VES对MCF-7乳腺癌细胞三苯氧胺的敏感性具有增强作用,可能与降低HER-2基因表达产物有关。 Objective To investigate the function of vitamin E succinate on the sensitivity of MCF-7 human breast cancer cells to tamoxifen. Methods MCF-7 human breast cancer cells were treated with VES for 24h at the concentrations of 5μg/ml and 10μg/ml. Then tamoxifen was administrated at the concentrations of 1μM, 2.5μM, 5μM and 10μM for 24h. The inhibitory effect was measured with MTT method. RT-PCR was applied to detect the transcript of HER-2 gene. Product of HER-2, c-erbB-2 protein expression in cancer cells was detected by immunohistochemistry. Results VES could improve the sensitivity of breast cancer cells to tamoxifen treatment. Both HER-2 transcript and c-erbB-2 protein expression were down regulated by VES. Conclusion VES can enhance sensitivity to tamoxifen in breast cancer cells. The mechanism maybe relate to the down regulation of HER-2 gene.
