摘要
[目的]研究细胞色素C(cytochromeC,cyt-C)在铝致海马神经细胞凋亡中的变化,为进一步研究铝致神经细胞凋亡的作用机制提供一定的资料。[方法]健康成年雄性SD大鼠40只,用随机数字表法按体重随机分为4组:生理盐水组、2.5mg/kgAl3+组、5.0mg/kgAl3+组、10.0mg/kgAl3+组。腹腔注射AlCl3溶液染毒60d后,处死,①用TUNEL法检测海马神经细胞凋亡;②Western-blot法检测海马神经细胞中cyt-C的表达情况,凝胶成像系统和扫描仪采集图像并对结果进行分析。[结果]①与对照组相比,Al3+5.0、10.0mg/kg组凋亡指数显著升高(P<0.05),Al3+5.0、10.0mg/kg组比Al3+2.5mg/kg组凋亡指数显著升高(P<0.05),Al3+10.0mg/kg组比Al3+5.0mg/kg组凋亡指数显著升高(P<0.05)。②与对照组相比,Al3+5.0、10.0mg/kg组cyt-C表达升高,差异有统计学意义(P<0.05)。海马神经细胞凋亡指数与cyt-C呈正相关(r=0.616,0.574,P<0.01)。[结论]铝诱导大鼠海马神经细胞凋亡可能与cyt-C释放入胞质有关。
[ Objective ] To explore the role of cyt-C in aluminum-induced the apoptosis of neurocytes in rats' hippocampus. [ Methods ] Fourty male SD rats, adult and healthy, were divided into 4 groups at random according to their weight using the random digital list method : normal saline, Al^3+2.5 mg/kg, Al^3+5.0 mg/kg, Al^3+ 10.0 mg/kg, They were exposed to AICI, by intraperitoneal injection for 60 d. After exposure, ①The apoptosis of neurocytes in rats' hippocampus were measured by methods of TUNEL. ②cyt-C in cytoplasm was detected by the method of Western-blot, the images were collected respectively by a gelatum analyzing system and scanner. The resuhs were analyzed by the gelatmn analyzing system. [ Results ] ① The apoptosis index ( AI )of neurocytes in rats' hippocampus in Al^3+ 5.0 mg/kg group and Al^3+ 10.0 mg/kg group was significantly higher than that in the control group ( P 〈 0.05 ), and AI in Al^3+10.0 mg/kg group was significantly higher than that in Al^3+2.5 mg/kg group ( P 〈 0.05 ). ② The expression of eyt-C significantly increased in the neurocytcs in rats' hippocampus in Al^3+5.0 mg/kg group and Al^3+10.0 mg/kg group compared with the control group ( P 〈 0.05 ). [ Conclusion ] Aluminum-induced apoptosis of neurocytes in rats' hippocampus is related with the release of cyt-C,
