摘要
目的探讨βcatenin在胃癌组织细胞核和膜/质中的含量与细胞增殖、浸润的关系。方法采用ELISA法测定βcatenin在胃癌细胞核或细胞膜/质中的含量,用免疫组化二步法检测cyclinD1、cmyc和Ki67蛋白在胃癌组织中的表达。结果在27例胃癌中细胞核与细胞膜/质中βcatenin含量之比与肿瘤的浸润深度呈正相关(rs=0.39225,P=0.0430),而与胃癌的Lauren分型无关;胃癌细胞核中βcatenin含量与cmyc蛋白或cyclinD1蛋白阳性表达呈正相关(rs=0.50904,P=0.0067;rs=0.39992,P=0.0387),细胞膜/质中βcatenin含量与cyclinD1蛋白表达亦呈正相关(rs=0.43612,P=0.0230)。但胃癌细胞核中βcatenin的含量、cyclinD1和cmyc蛋白的阳性表达与Ki67均无相关性。结论胃癌细胞核与癌细胞膜/质中βcatenin含量之比与胃癌的浸润性具有相关性,而与胃癌的Lauren分型无直接的相关性。βcatenin在细胞核内的积聚可促进cyclinD1和cmyc的表达,有利于胃癌细胞增殖。
Purpose To investigate the relationship of the quantity of β-catenin in nuclei and cytoplasm with the cellular proliferation and infiltration on gastric carcinoma. Methods The quantity of β-catenin in nuclei and cytoplasm were analysed by ELISA;The expressions of cyclin D1, c-myc and Ki-67 were examined by immunohistochemistry (IHC). Results In 27 cases of gastric carcinoma, the ratio of β-catenin content between nuclei and cytoplasm was positively related to depth of infiltration ( rs = 0. 392 25, P = 0. 043 0), the content of β-catenin in nuclei was positively related to the expression of c-myc (rs = 0. 509 04, P = 0. 006 7 ), and also to the expression of cyclin D1 ( rs = 0. 399 92, P = 0. 038 7 ). The content of β-catenin in cytoplasm was also positively related to the expression of cyclin D1. However, the content of β-catenin in nuclei and the positive expression of cyclin D1 and c-myc had no relation with Ki-67. Conclusions The ratio of the content of β-catenin in nuclei to that of cytoplasm is closely correlated with tumoral infiltration, but not directly related to the Lauren classification of gastric carcinoma. The accumulation of β-catenin in nuclei might stimulate the expression of cyclin D1 and c-myc, and hence facilitate the proliferation of the tumor cells.
