Influence of focal adhesion kinase on the biological behaviors of human glioma cells

摘要 目的探讨反义黏着斑激酶(FAK)对U251人胶质瘤细胞株生物学行为影响。方法以LipofecTAMINE^(TM)介导的反义FAK寡核苷酸转染U251人胶质瘤细胞株,用Western blot检测胶质瘤细胞株FAK含量的改变,研究其多种细胞生物学行为的变化。结果脂质体介导FAK ODN转染率为85.9%。转染反义FAK后,U251人胶质瘤细胞株FAK表达明显减少,L1251细胞生长抑制率为64.52%,侵袭细胞下降至21.7个,细胞周期分析显示s期细胞降至25.16%,细胞凋亡率升至34.5%。透射电镜观察到凋亡细胞。结论反义FAK寡核苷酸可显著降低FAK在U251 中的表达,降低FAK表达能够显著抑制U251人胶质瘤细胞株侵袭能力,抑制细胞增殖和诱导凋亡。 Objectives To study the effect ol antisense focal adhesion kinase (FAK) oligodeoxynucleotide on the biological behaviors of U251 MG cells. Methods LipofecTAMINE-mediated antisense FAK ODN was transfected into U251 MG ceils. Western blot analysis was used to detect the expression of FAK. The biological behaviors of human glioma cells were investigated. Results FAK antisense ODNs were transfected efficiently by cationic lipid (85.9 % ). FAK were inhibited significantly after FAK antisense ODNs transfection. For U251 MG ceils, antisense FAK ODNs significantly inhibited the proliferation by 64.52% and reduced the invasion cell number to 21.7. The ceils at S phase were decreased to 25.16 % and the occurrence of apoptosis increased significantly to 34.5 % in the group treated with antisense FAK ODNs. Apoptosis ceils were observed by transmission electron microscopy. Conclusion Antisense FAK ODNs can decrease the FAK expression significantly, resulting the reduced ability of U251 MG cells to invade. FAK suppression significantly inhibits the proliferation of U251MG cells and induces their apoptosis.
