摘要
探讨大鼠脑缺血再灌注损伤(MCAO)后内源性神经干细胞的增殖分化情况,以及大鼠脑缺血再灌注损伤(MCAO)后血浆雌二醇(E2)含量变化和通心络对其影响。方法:采用大鼠缺血再灌注损伤(MCAO)模型,应用放免方法观察缺血后3天、7天、14天以及21天E2含量变化,免疫组织化学方法观察缺血后缺血侧室管膜及室管膜下区(SVZ)、海马齿状回(SGZ)BrdU阳性细胞数的变化。结果:造模后通心络组BrdU阳性细胞荧光强度值明显高于脑缺血再灌注模型组,差异显著(P<0.05);MCAO血浆E2浓度显著降低(P<0.05),通心络能升高E2浓度。结论:大鼠缺血再灌注损伤后可引起其缺血侧SVZ、SGZ区星形胶质细胞及神经细胞反应和增殖;而通心络可显著增加MCAO大鼠神经干细胞增殖分化能力,E2可能起着重要作用。
Objective : To investigate the change of estradiol (E2) and the differentiation of neural stem cells after middle cerebral artery occlusion in rats and treatment with Tongxinluo. Methods: Ischemia was induced by temporary middle cerebral artery occlusion (MCAO).At 3, 7, 14 and 21 days after MCAO, the amounts of E2 was detected, In ependyma and subependymal region (SVZ), Hippocampos dentate gyrus (SGZ), the numbers of BrdU was detected by immunohistochemistry. Results: Compared with the sham operation group, E2 was significantly decreased in the MCAO on the day 74 14 and 21 (P 〈 0.05) .After treatment of Tongxinluo, the amounts of E2 had significantly increased in the MCAO; the number of BrdU was significantly increased as compared with the MCAO group ( P 〈 0.05) o Conclusions : Tltis study demonstrates that tbcal cerebral isellemia in the rat results in a rapid response, from resident astrocytes and nerve cell of the SVZ and SGZ to the side of ischenlia, Tongxinluo enhances the ability of the differentiation and pro liferation of neural stem cell in MCAO; E2 might play an important role in cerebral isehemic injury.
