罗红霉素对哮喘儿童外周血单个核细胞Th1/Th2平衡的影响 被引量:4

摘要 目的观察不同浓度罗红霉素对哮喘儿童外周血单个核细胞(PBMC)中两个功能性亚群(TH1/TH2)功能状态的影响。方法采用酶联免疫吸附法(ELISA)检测20例哮喘及19名健康儿童PBMC经不同浓度的罗红霉素体外干预培养后,上清中的IFN-γI、L-4I、L-10和IL-12水平变化。结果哮喘组儿童的PBMC分泌IL-4明显增多,IFN-γ/IL-4比值明显降低,哮喘组50μg/mL的罗红霉素可明显促进哮喘儿童PBMC分泌IFN-γ和IL-12,并可恢复TH1/TH2平衡,亦有促进IL-10分泌的倾向。结论50μg/mL的罗红霉素可明显促进IFN-γ及IL-12分泌,恢复哮喘儿童体内TH1/TH2平衡,并能促进IL-10分泌,有利于协调机体细胞和体液免疫的功能。 Objective To observe the effects of roxithromycin (RXM) on the expression of Th1/Th2 cytokine in peripheral blood mononuclear cells(PBMCs) of asthmatic children. Methods Twenty asthmatic children and 19 healthy children were included. PBMCs were isolated from venous blood. Fifty mg/L or 5 mg/L RXM was added to cultures at the time of initial stimulation. The levels of imerferon-γ( IFN-γ), interleukin(IL) -4, IL- 10 and IL- 12 in culture supematants were determined by enzyme- linked irrnnunosorbent assay. Results Fifty mg/L RXM could promote the secretion of IFN -γ obviously and IL- 12, but not IL- 4. It also tended to increase the production of IL- 10. Fifty nag/L RXM could restore the imbalance of Th1/Th2 cell function in terms of IFN-/IL -4 bioactivity, Conclusions Fifty mg/L RXM may promote the secretion of IFN -γ and IL- 12 obviously, and re.store the balance of Th1 / Th2 existed in asthmatic children ,and induce IL- 10 secretion as well. RXM may be used in clinical treatment of asthma.
