摘要
目的:研究实验性大鼠肝脏纤维化进程中瘦素表达的变化。方法:雄性W istar大鼠23只,随机分成2组。正常对照组不做处理;四氯化碳造模组,40%四氯化碳蓖麻油溶液皮下注射(3m l/kg,每周2次,首剂加倍)。分别于第4,8周取材。检测血清肝功能、肝纤维化指标和肝脏中瘦素表达的变化,光镜下观察组织学的改变。结果:模型组血清肝功能、肝纤维化、血清瘦素和肝脏中瘦素表达都加重,与对照组比较有统计学差异。模型组中血清瘦素和肝脏中瘦素的表达在8周末与4周末之间有统计学差异。结论:实验性大鼠肝纤维化时明显有瘦素的表达,并且随肝纤维化程度的加重而增加。
Objective: To study the leptin expression in rat hepatic fibrosis. Methods: 23 male Wistar rats weighting about 230 - 270 g were randomly divided into two study groups : normal group and CCl4 model group. The experimental hepatic fibrosis was induced by subcutaneous injection of carbon tetrachloride oil solution. In the 4^th and 8^th weeks of the experiment, rat liver tissues were taken for morphological analysis (by FIE stained sections) and the stage of hepatic fibrosis was observed under light microscope. Blood sample were collected to measure serum transaminases ( AST and ALT) and ALB. Serum procollagen ⅢN-terminal peptide (PⅢNP) ,hyaluronic acid(HA) and leptin were measured using radio-immunity technique. The protein expressions of leptin in liver tissue were determined with immunohistochemistry assay. Results: Morphological and biochemistry analysis showed that the liver function worsened and the liver fibrosis was exacerbated in the CCl4 induced rats, while the expression of leptin was increased in liver and serum of the model group, compared with those in normal group ( P 〈 0. 05 or P 〈 0. 01 ) . There were significant differences between the liver and serum leptin levels at the 4^th and the 8^th weeks. Conclusion: leptin are involved in the formation and development of liver fibrosis.
