摘要
目的:探索一种简便可重复的脑缺血动物模型建立方法。方法:选用250~300g SD成年大鼠20只,采用改良的Longa线栓法制作大鼠局灶性脑缺血模型,并评价缺血效果。结杲:术后大鼠均出现了偏瘫等神经功能障碍,镜下观察可见大鼠大脑左侧额顶皮质以及基底节区大范围的梗死区,呈现白色。结论:神经功能损害以及1% TTC大鼠脑切片染色显示改良线栓法建立局灶性脑缺血模型效果较好。
Objective. To explore a reliable method of focal cerebral ischemia in rats. Methods: Focal cerebral ischemia model was made by reforming Longa method in Sprague-Dawley rats and neurological deficit of all rats were graded after suture occluded. Internal carotid artery and common carotid artery were tied with suture which was retracted by forceps in order to reduce the bleeding in process. External artery was ligated and cut 2mm long before the beginning of bifurcation of common carotid artery. The obstruction of the middle cerebral artery was performed by inserting 0.2mm monofilament nylon suture in the left internal caroted artery via external carotid artery until it reach and occlude the ostium of the left middle cerebral artery. Result: All rats were made semi paralyzed after procedure. Conclusion. It is a reliable way to made focal ischemic model by reforming Longa method that focal ischemic lesion well set up with neurological deficit in rats.
