基于COⅠ基因部分序列对尾凤蝶属(鳞翅目,凤蝶科)四种蝴蝶分子系统关系及相关问题的探讨 被引量:34

MOLECULAR PHYLOGENETIC RELATIONSHIPS AMONG FOUR SPECIES OF BHUTANITIS (LEPIDOPTERA, PAPILIONDAE) BASED ON PARTIAL COⅠ GENE SEQUENCE

摘要 为了探讨中国尾凤蝶属的系统发生关系,对中国产4种尾凤蝶属蝴蝶的细胞色素氧化酶Ⅰ(COⅠ)基因的部分序列进行了分析。在获得的636bp的序列中,有59个变异位点,47个简约信息位点;A、T、G、C碱基的平均含量分别为29.5%、40.8%、13.9%及15.8%;表现出明显的A+T含量(70.3%)偏异。采用NJ、MP、ML法构建了分子系统树,结果表明尾凤蝶属二尾种组与多尾凤蝶亲缘关系较近,而三尾凤蝶较早分化出来。同时结果表明丽斑尾凤蝶和二尾凤蝶的COⅠ基因没有序列差异。对这2个物种3个个体的COⅡ基因进行了测序,结果也没有序列的差异,表明丽斑尾凤蝶和二尾凤蝶可能是同物异名。而三尾凤蝶的遗传多样性较为贫乏。 The phylogenetic relationships among 4 Bhutanitis species from China were analyzed based on partial OD Ⅰ gene sequence (about 636 bp). There were 59 variable sites and 47 parsimony-informative sites in the 636 nucleotides of the data, Bhutanitis exhibits a 70.3% A+ T bias. Molecular phylogenetic trees were reconstructed using Neighbor-joining method (N J), Maximum Parsimony (MP) method and Maximum Likelihood (ML) method. The molecular phylogenetic trees showed that mansfieldi group plus Bh. lidderdalii is the sister group of Bh. thaidina. On the other hand, there was no sequence divergence between CO Ⅰ gene sequence of Bh. mansfieldi and Bh. Pulchristata. Partial CO Ⅱ gene sequences from the 3 individuals of the 2 species were also sequenced and analyzed, there was no sequence divergence either. The results indicated that Bh. pulchristata should be treated as a synonym of Bh. mansfieldi. The results also showed that the mtDNA divergence of Bh. thaidin, was lower.
