SCE诱导人结肠癌细胞凋亡及其机制研究

摘要 【目的】观察鲨鱼软骨提取物(SCE)对人结肠癌细胞(THC-8908)凋亡的诱导作用,并进一步研究在此过程中Bcl-2和Caspase-3的表达。【方法】将不同浓度的SCE作用于THC-8908细胞,观察其作用的时间效应及剂量效应;在光镜和电镜下观察其形态变化;用免疫细胞化学法观察Bcl-2和Caspase-3的表达。【结果】SCE能抑制THC-8908细胞生长,在一定剂量和时间范围内引起细胞凋亡,并显示剂量和时间效应,在此过程中Bcl-2蛋白表达减弱,Caspase-3表达增强。【结论】SCE能明显抑制THC-8908细胞体外生长,并可诱导其细胞凋亡,其作用机制可能与下调Bcl-2蛋白的表达并上调Caspase-3的表达有关。 [Objective] To observe the effect of shark cartilage extract (SCE) on apoptosis and the express of Bcl-2 and Caspase-3 in Human colon cancer cell line (THC-8908). [Methods] Different concentrations of SCE were used to treat THC-8908 ceils and inhibition of proliferation was measured by MTT assay. Cell morphology was observed under light and electron microscope. The expression of Bcl-2 and Caspase-3 were studied by immunocytochemical technique. [Results] SCE could inhibit the growth of THC-8908 cell. Within a certain range of treating time and dose, apoptosis were induced with a time and dose related manner. The expression of Caspase-3 was increased and that of Bcl-2 was decreased (P 〈 0.01 ). [Coneluslon] SCE could inhibit growth of THC-8908 cells significantly and induce apoptosis of THC-8908 cells, and decrease of Bcl-2 and increase of Caspase-3 may play an important role in the process.
