摘要
目的检测妊娠早期子宫微环境中蜕膜、滋养层及子宫NK(uNK)细胞基因表达变化,为揭示妊娠中母胎免疫屏障及胚胎发育与植入等过程中的生理与病理机制提供新的研究靶点。方法首先用免疫磁珠(MACS)技术分离及纯化uNK细胞,流式细胞仪检测其纯度,RT-PCR技术检测与uNK细胞的侵袭、促血管形成等生物学活性有关基因的转录水平,同时检测蜕膜与滋养层组织相应的配体表达状况。结果通过MACS技术检测,所获得的NK细胞纯度较高,大于95%,RT-PCR检测发现妊娠早期uNK细胞可表达多种与侵袭和血管形成等功能有关的生物活性分子,同时在蜕膜与滋养层组织中检测到相应受体的表达。结论妊娠早期uNK细胞是一群具有强大调节功能的NK细胞亚群,在调控滋养层细胞的侵袭、母胎界面血管重建与免疫耐受形成中可能起重要的作用。
Objective Studying uNK cells(uterine natural killer cells), decidual and trophoblast tissue gene expression for offering, a new research target in the pregnancy physiology and pregnancy pathology. Methods Microbeads were used to isolate and purify uNK cells. FACS analyzed purity. RT-PCR technique was applied for the proving evidence d the invasion, angiogenesis and other biological activity of uNK cells, at the same time, detecting ligand expression on decidual tissue and trophoblast tissue. Results uNK cells express some gene which relate to invasion and angiopoiesis. At the same time, decidual tissue and trophoblast tissue expressed their ligands. Conclusion uNK cells function as one substantial regulative NK subset. They contribute to monitor the invasion of trophoblast cells and maintain vascular modification and immune tolerance at fetal-maternal interface.
