GFRα1蛋白在小鼠隐睾精原细胞中的表达及意义

摘要 目的探讨胶质细胞源性神经营养因子受体α(GFRα1)蛋白在小鼠隐睾精原细胞中的表达及对精原干细胞增殖的调节作用。方法取成年小鼠40只制作单侧隐睾模型,饲养2~3个月后,切取睾丸行光镜和电镜观察,并用免疫组织化学方法检测睾丸细胞GFRα1的表达,与对侧睾丸做对照。结果隐睾模型GFRα1主要表达在位于基底膜的As型精原干细胞,在Apr型细胞和Aal型细胞未见表达,与对侧相比。GFRα1表达增多.P〈0.05。结论隐皋睾丸组织中Aal→A1分化受阻,已分化的精原细胞缺失,反馈性的GFRα1表达增多.可促进精原干细胞增殖。 Objective: To discuss expression of glial cell-derived neurotrophic factor (GDNF) receptorα1 protein (GFRα1)in the germ cells of eryptorchid mice and regulating proliferation of germinal stem cell (GSC). Method: 40 unilateral artificial cryptorchidism models were established with 6-8 weeks old male KM mice. 2-3 months after operation, testis were cut and observed by light and electron microscopy. GFRα1 expression in testis tissue was detected by immunohistochemnistry. The opposite side testis were used as control. Result: GFRα1 was mainly expressed at As germinal stem cells near to basement membrane, not expressed at Apr and Aal. Its expression increased compared to the control (P 〈 0. 05). conclusion: In cryptorchid testis, the differentiation from Aal to A1 was blocked. The differentiated germ cells depleted. GFRα1 expressed increasely by feedback mechanism and promoted the proliferation of GSC.
