摘要
目的提高教学质量,探讨如何增强异染颗粒染色,提高标本的清晰度。方法按常规接种白喉杆菌于甘油鸡蛋或吕氏培养基上。用传统的奈瑟法(Neisser)及改进法(吕氏美兰+俾褐麦斯),染色步骤、时间按常规。用普通显微镜观察两种方法每张标本的染色情况及异染颗粒。用德国KONTRONIBAS2.0全自动图像分析仪随机抽查两种方法染色标本的异染颗粒数目。结果传统奈瑟法的异染颗粒数目相对少些,着色浅。改进法染色的标本涂片均可见明显异染颗粒,数目相对比传统奈瑟法多,着色深。结论用改进的奈瑟(Neisser)染色法染色白喉棒状杆菌,异染颗粒数量多,染色效果优于传统奈瑟法。
Objective To study how to enhance the staining of the metachromatic granules of C.diphtheriae, and to increases teaching quantity. Methods The samples were prepared by the inoculation and cultivation as usually. One half of the sample was stained by the traditional Neisser stain and the others by the improved method. Result The metachromatic granules of the samples stained by the improved method were more than those stained by the traditional method. Conclusion The improved Neisser stain is better than the traditional one.
