摘要
目的探讨过量谷氨酸钠对豚鼠视网膜内突触小泡蛋白表达的影响及其意义。方法实验组给予腹腔注射谷氨酸钠3g·kg-1·d-1,连续7d,对照组注射等量生理盐水,分别在注射后10天取材。采用免疫组织化学方法和图像分析技术,对两组豚鼠视网膜内突触小泡蛋白的免疫反应产物进行检测。结果对照组豚鼠视网膜内突触小泡蛋白的表达为强阳性,主要定位在外网状层和内网状层,实验组视网膜相应区域内阳性反应产物表达的光密度值明显低于对照组(P<0.01)。结论过量谷氨酸可明显抑制视网膜突触小泡蛋白的表达,这可能是造成视觉功能障碍的重要原因。
Objective: To investigate the effect and significance of superabundant glutamate on expression of synaptophysin in guinea pig retina, Methods. Guinea pigs were divided into experimented group , which was given ghutamate at 3g·kg^-1·d^-1ip, for 7 d, and control group that accepted equivalent normal saline, After 10 days after injection, the expression of synaptophysin in retina was detected with immunohistochemical merhod and image analysis, Results. Synaptophysin in retina of control group showed strong positive reaction. The molecule was expre.ssed mainly in inner and outer ptexiform layers of the retina. The optical density of synaptophysin immunoreactivity in retina of expermental animals decreased significantly compared with controls ( P 〈0. 01). Conclusion. The superabundant glutamate can obviously inhibit expression of synaptophysin in the retina, and this probably was a main realign resulting in dysfunction of sight.
