摘要
目的探讨真核细胞翻译起始因子4E(EIF4E)和非霍奇金淋巴瘤(NHL)血管生成的关系。方法检测52例初诊NHL患者淋巴瘤石蜡切片EIF4E和平均微血管密度(MVD)的表达。构建反义EIF4E表达载体,电转染至Burkirt淋巴瘤细胞系Raji细胞,应用western-blot方法检测Raji细胞转染前后和正常人外周血淋巴细胞(NPBMC)中血管内皮生长因子(VEGF)的表达。结果在NHL患者中,侵袭性淋巴瘤EIF4E和MVD的表达明显高于惰性淋巴瘤的表达(P<0.05),且EIF4E与MVD的表达呈正相关(r=0.695,P<0.01)。成功构建EIF4E反义表达载体,将其转染入Raji细胞后,VEGF表达抑制。结论EIF4E在NHL的肿瘤血管生成中发挥作用,可能通过上调VEGF的表达而促进肿瘤血管生成。
Objective To study the relationship between eukaryocytic translation initiation factor 4E (EIF4E) and MVD in angiogenesis ofnon-Hodgkin lymphoma. Methods Mean microvessel density (MVD) and EIF4E were detected in 52 lymph node samples paraffin sections of patients with newly diagnosed NHL by the way of immunohistochemistry. Antisense EIF4E cDNA was clone into plasmid pcDNA3.1(+) and constructed plasmid was transfected into Raji cells. EIF4E protein expression and vascular endothelial growth factor (VEGF) proteins were detected by Western blot in untreated Raji, plasmid transfected Raji cells and normal human peripheral blood monocular cells (NPBMC). Results Expressions of EIF4E and MVD were higher in aggressive lymphomas than in indolent lymphomas(P〈0.05). The expression of EIF4E was positive correlated with MVD in lymph node of NHL (r = 0.695, P〈0.01). Antisense EIF4E eukaryotic expression vector (pcDNA3.1-EIF4Eas) was constructed successfully. After blockage with EIF4E by pcDNA3.1-EIF4Eas in Raji cells, EIF4E and VEGF decreased expressions. Conclusion In NHL, EIF4E expression is correlative with MVD on angiogenesis. EIF4E play an important role in NHL tumor angiogenesis and can regulate the protein level of VEGF in non-Hodgkin lymphoma cell lines Raji.
