摘要
目的建立一种检测金黄色葡萄球菌A型肠毒素的PCR方法,以期用于葡萄球菌肠毒素食物中毒的诊断。方法根据文献报道选择金葡菌SEA基因中比较保守的片段设计引物,优化各种工作条件,建立PCR检测方法,并鉴定其敏感性和特异性以及抗杂菌干扰性。扩增片段应用限制性核酸内切酶RsAI消化进行分析。结果所建立的方法敏感性和特异性均较好,有较强的抗杂菌干扰能力。结论应用PCR法检测SEA基因为SEA基因的诊断提供了病因学基础。
Objevtive To development a PCR assay for detecting staphylococcus aureus Enterotoxin A (SEA). Methods According to the sequences of conservative areas of the SEA gene, primers were selected and used to amplify SEA gene. The amplified fragments were analyzed by digestion of restriction endonuclease RSAI. Result The established assay has high sensitivity and good specificity, anti-interference to other bacterial is satisfied. Conclusion The PCR assay for detecting SEA provided an etiological basis for the diagnosis of SEA gene.
