灯盏花素对异丙肾上腺素诱导大鼠实验性心肌肥厚的影响 被引量:4

摘要 目的研究灯盏花素(Breviscapine)对异丙肾上腺素(Isoproterenol)引起大鼠实验性心肌肥厚的保护作用。方法异丙肾上腺素皮下注射连续7天建立大鼠心肌肥厚模型。造模第2天起腹腔注射灯盏花素12.5和25mg/(kg·d),连续用药14天,测量大鼠心脏重量指数(HW/BW)和左心室重量指数(LVW/BW),分光光度法检测左心室心肌组织中羟脯氨酸、胶原蛋白含量和Na+,K+-ATPase,Ca2+-ATPase活性。结果异丙肾上腺素模型组大鼠心重指数和左心室重量指数明显增大,左心室心肌组织中羟脯氨酸和含量胶原蛋白增高,Na+,K+-ATPase和Ca2+-ATPase活力下降;灯盏花素能显著增强Na+,K+-ATPase和Ca2+-ATPase活力,降低羟脯氨酸含量,抑制胶原的产生。结论灯盏花素对对异丙肾上腺素引起大鼠实验性心肌肥厚具有一定的改善作用。 Objective: To investigate the protective effects of breviscapin on myocardial hypertrophy induced by isoproterenol in rats. Methods : Myocardial hypertrophy model of rats were induced by isoproterenol (5 mg/kg · d sc) for 7 days. From day 2 - day 15 ,rats were treated with breviscapin 12.5 and 25 mg/kg · dip ,the NS control and isoproterenol model group received saline injection. On day 15, animals were killed by decapitation, monitored the myocardial indexes ( heart weight/boday weight, HW/BW and left ventricalar weight/body weight, LVW/BW) ;the contents of hydroxyproline, collagen and the activity of Na^ + , K ^+-ATPase and Ca^2+-ATPas in left ventricle were assayed by spectrophotometry. Results : Compared with NS control group, the myocardial indexes, the level of hydroxyproline , collagen in left ventricle were markedly increased , activity of Na ^+ , K ^+ -ATPase, Ca^2+ -ATPas in left ventricle were decreased in isoproterenol model group;treatment with breviscapin 25 mg/kg · d significantly reduced the myocardial index and content of hydroxyproline and collagen in left ventricle, enhanced the activity of Na ^+ , K ^+ -ATPase and Ca^2 + -ATPas. Conclusion Breviscapin could alleviate myocardial hypertrophy induced by isoproterenol in rats.
