桑枝、桑白皮体外降糖作用研究

Study on hypoglycemic effects of extracts of Ramulus Mori(桑枝) and Cortex Mori (桑白皮)in vitro

目的研究桑枝、桑白皮体外降糖作用,初步探讨其降糖作用机理。方法采用与人肝细胞表型相似的HepG2细胞,检测24h后培养液中葡萄糖的消耗量,用MTT法监测细胞增殖的情况。结果桑枝、桑白皮在高糖状态下可使HepG2细胞的葡萄糖消耗量增加,对胰岛素刺激的HepG2细胞的葡萄糖消耗量增加有协同作用。结论促进外周组织的葡萄糖代谢、提高肝细胞对胰岛素的敏感性可能是桑枝、桑白皮防治2型糖尿病作用的机理之一。

Objectsive ： To observe the effects of extracts of Ramulus Mori and Cortex Mori on glucose uptaking in cuhured HepG2 cells in vitro and investigate the hypoglycemic mechanism of them initially. Methods： HepG2 cells , the phenotype of which is similar to that of the human hepatoeytes , were used to detect the 24 hours, consumption of glucose in cultured fluid , MTT assay was used to monitor the proliferation of the HepG2 cells. Results： Ramulus Mori and Cortex Mori could increase the glucose consumption of HepG2 ceils and enhance the glucose consumption in coordination with insulin. Conclusions： Promoting the glucose uptake rate of peripheral tissue and enhance the sensitivity of hepatocytes to insulin may be one of the possible hypoglycemic mechanisms of Ramulus Mori and Cortex Mori.
