摘要
目的观察17β-雌二醇(E2)对脾细胞的影响,分析子宫内膜异位症雌激素-免疫的病理环节。方法MTT法检测不同浓度雌二醇对静息脾细胞、ConA活化脾细胞增殖的影响、雌激素受体拮抗剂Tamoxifen对E2作用的影响及桑寄生对E2的干预。结果雌二醇对静息脾细胞增殖未见明显影响,10-6ME2明显抑制2.5μg/ml ConA活化的脾细胞增殖,Tamoxifen可以抑制E2对ConA活化的脾细胞的增殖抑制作用,桑寄生也有相同的作用。结论高浓度的雌二醇可以抑制脾细胞的增殖,此作用可以被雌激素受体拮抗剂拮抗,提示高浓度的E2可以抑制免疫细胞的活化,桑寄生拮抗E2抑制ConA活化的脾细胞增殖的作用。
Objective: To investigate the effect of estradiol on splenocytes in vitro, to probe the immune clue of the pathogenesis of endometriosis. Method: By using the MTY , the proliferation of naive and activated splenocytes ( by concanavalin A ) were measured under different level of estradiol by MTY. The intervencing effect of Tamoxifen and Sangjisbeng on estradiol -splenocytes were also reasearched by MTT. Result: Fatradiol has no obvious effect on the number of naive splenocytes rang from 10^-10-10^-6 M. But the proliferation of activated splenocytes has been obviously inhibited while treated with 10^-6M estradiol. This inhibition of estradiol can be alleviated significantly by Tamoxifen and Sangjisheng. Conclusion: Proliferation of activated splenocytes is inhibited at the high level of estradiol ,which can be alleviated by antago- nist of estradiol receptor modulator Tamoxifen and Sangjisheng. It suggested that the mitogen actived splenocytes may more susceptible to the E2 , and the E2 receptor on the immune ceils play a key role in the communication between Ez and splenocytes, because the Tamoxifen can alleviate the proliferation of splenocytes. The phenomena may be a immune clue for probing the pathogenesis of endometriosis. The further study is needed. As for the Sangjisheng,the traditional Chinese medicine, its alleviated the E2 inhibition mitogen actived splenocytes has not been demonstrated, and the mechanism is very interesting for us to observe.
