胸腺素Α_1对支气管哮喘患者气道高反应的抑制作用 被引量:1

摘要 目的探讨胸腺素Α1(TΑ1)对哮喘患者TH1/TH2免疫反应的调节以及其对气道高反应性的抑制作用。方法选取缓解期哮喘患者25例(哮喘组),给予胸腺素Α1(TΑ1)隔日一次皮下注射,每次1 MG/KG,疗程1.5月。健康志愿者(对照组)15名。分别对哮喘组用药前、后,对照组留取血标本,采用双抗体夹心酶联免疫吸附法(ELISA)检测上述三组血清中IL-4、IL-5、IFN-Γ含量,并对检测结果进行统计学处理。结果哮喘组用药前血清中IL-4含量[(52.15±8.96)PG/ML]与对照组[(13.33±11.16)PG/ML]比较差异有显著性(P<0.01)。哮喘组用药后血清IFN-Γ量[(1185.52±67.10)PG/ML]显著增加,与对照组[(138.25±6.25)PG/ML]和用药前[(144.58±1.06)PG/ML]相比均有显著性差异(P=0.001);IL-4含量[(14.15±4.86)PG/ML]较用药前[(52.15±8.96)PG/ML]明显降低(P=0.001)。结论本研究显示:TΑ1能上调TH1型免疫反应,下调TH2型免疫反应,抑制IL-4和IL-5的过度表达,从而对哮喘患者的气道高反应性有治疗和抑制作用。 Objective To investigate the role of thymosin-α1 ( Tα1 ) administrated Th1/Th2 balance and suppression of allergic sensitization in asthmatics. Methods Twenty-five asthmatic patients in remission episode were selected ( the asthma group). Fifteen healthy volunteers ( the control group) was selected. The asthma group was hyodermiced by Tα1 every other day for 1.5 months. Serum were obtained from the asthma group before and after treatment and from the control group. IL-4, IL-5, and IFN-γ levels in above-mentioned serum samples were measured by ELISA. The results were analyed statistically. Results IL-4 levels of asthma before treatment [ (52. 15 ± 8.96 ) pg/mL] were significantly increased compared with those of control group [ ( 13.33 ±11.16 ) pg/mL] ( P 〈 0. 05 ). IFN-γlevel in asthmatics after treatment [ ( 1185.52± 67. 10) pg/mL ] was more significantly increased than that in control group [ ( 138.25 ± 6. 25)pg/mL] and that of asthmatics before treatment[ ( 144. 58 ±1.06) pg/mL] (P = 0. 001 ). IL-4 Levels in asthmatics after treatment [ ( 14. 15 ±4. 86)pg/mL] were more significantly decreased than those in asthmatics before treatment [ (52. 15 ±8.96) pg/mL] ( P=0. 001 ). Conclusion In this study,Tα1 treatment established a Th1-type immune response that thindered allergic sensitization and the development of increased airway reactivity.
