摘要 目的观察单核细胞趋化蛋白1(MCP1)对体外培养的大鼠系膜细胞增殖及纤维连接蛋白(FN)和Ⅰ型胶原(COLⅠ)基因表达的影响。方法采用不同浓度的MCP1刺激体外培养的系膜细胞,于不同的时间应用MTT法测定系膜细胞增殖活性,半定量RTPCR的方法测定细胞FN和COLⅠmRNA的相对表达量。结果随着MCP1浓度的增高,刺激时间的延长,MCP1明显促进系膜细胞的增殖、上调细胞FN和COLⅠmRNA的表达(Ρ<0.01),表现为剂量和时间依赖性。结论MCP1能促进大鼠系膜细胞的增殖及上调细胞FN和COLⅠmRNA的表达。MCP1可能在系膜增生性肾小球肾炎的发病过程中起一定的作用。 Objective To explore the effect of monocyte chemoattractant protein-1 (MCP-1) on cell proliferation and the mRNA expression of fibronectin(FN), collagen type Ⅰ (COL Ⅰ ) in cultured rat mesangial cells(MCs). Methods Rat MCs were cultured in vitro and incubated with different concentrations of MCP-1 in different time. The cell proliferative activity was assessed by MTT assay. Semi-quantitive RT-PCR was used to detect the mRNA expression of FN and COL I in MCs. Results MCP-1 could significantly stimulate the mesangial cell proliferation and remarkedly up-regulated the expression of FN and COL Ⅰ mRNA in a dose-and time-dependent manner (P〈0. 01). Conclusion MCP-1 could promote the cell proliferation and up-regulate the expression of FN and COL I mRNA in rat MCs cultured in vitro. MCP-1 play certain role in the pathogenisis of mesangial cell proliferative glomerulonephritis.
