摘要
SOX基因家族是一类与SRY基因高度相关的转录调控因子,具有一个保守基序HMG-box,可以与DNA序列特异性结合,促进基因的特异转录,从而在胚胎发育及性别分化中起着重要作用。本文采用PCR方法,扩增了凹耳蛙的SOX基因保守区,获得长约220bp的片段;SSCP分析结果显示凹耳蛙SOX基因雌雄个体片段无差异,与人的有较大差异,本文为探讨凹耳蛙的性别决定机制及SOX基因的进行提供了分子资料。
The SOX gene family of transcription factors highly related to the SRY gene. They are characterized by the presence of a HMG domain involved in various developmental processes. The encoded proteins are capable of binding to DNA in a sequence-specific manner. The technique of PCR was used in this paper to amplify the SOX gene of Rana tormotus with a pair of premiers which specially amplify the conservation sequences of man HMG-box gene. And analyze the amplification products with SSCP. There is only a band, about 220bp, in both male and female Rana tormotus. It suggested that the Sox gene of Rana tormotus is no difference between male and female Rana tormotus Wu, but have great difference comparing with that of human being. Our work supplies molecular data for the study for Sex-determining mechanism of Rana tormotus and for the evolution of Sox gene.
