苦丁茶中熊果酸含量的测定

Determination of Ursolic Acid Content in Ilex Kudincha C.J.Tseng

目的建立苦丁茶中熊果酸含量测定方法。方法采用高效液相色谱法测定苦丁茶中熊果酸含量,色谱柱:Diam onsil C18(4.6×250m m,5μm);流动相:乙腈-0.05%磷酸(92∶8);流速:1mL.min-1;柱温:30℃;检测波长:220nm。结果熊果酸在0.82～4.10μg范围内线性关系良好,平均回收率为98.2%,RSD=1.3%(n=5),苦丁茶嫩叶中熊果酸的含量为5.93mg/g,老叶中的含量为12.74mg/g。结论该方法简便可行、重复性好,可作为苦丁茶质量评价的依据。

Objective To establish a determinating method of ursolic acid in Ilex kudincha C.J.Tseng. Methods The content of the ursolic acid was detected with HPLC 220nm, Diamonsil C18 （4.6×250mm, 5μm） column, the mobile phase was a mixed liquid with Acetonitrile -0.05% H3PO4（92：8）. The column temperature was setup at 30℃ and the flow rate was 1 mL· min^-1 Results A good linearity of ursolic acid was in the range of 0.82-4.10μg, the average recovery was 98.2%, the content of ursolic acid in tender leaves on Ilex kudincha C.J.Tseng was 5.93mg/g, and the content of ursolic acid in older leaves was 12.74mg/g. Conclusions The method is rapid, sensitive and accurate. It can be used to control the quality of Ilex kudincha C.J.Tseng.
