细菌鞭毛染色法染色条件的探讨 被引量:5

摘要 目的 探讨不同的染色条件对细菌鞭毛染色效果影响,寻找一套适合的染色方法。方法 将37℃培养18-24h的普通变形杆菌培养物,稀释成菌悬液制片、染色,比较不同的菌悬液浓度、处理时间、染色时间和染色方法对细菌鞭毛染色效果的影响。结果浓度为1.5×10^12/U的菌悬液37℃放置10min制片,用石炭酸复红法染色10~15min,水洗,干燥镜检,其背景清晰,80%~90%的细菌有鞭毛,菌体红色,鞭毛淡红色,染色效果理想。结论 细菌鞭毛染色过程中菌悬液的浓度、菌悬液的处理时间、制片方法以及染色时间的长短等条件影响鞭毛染色效果。 Objective To study the staining of bacterial flagellum under different staining conditions and to find an appropriate way of it. Methods The Proteus vulgaris culture was cultured at 37℃ for 18-24 h, then a slide was made by the diluted suspension and stained. The staining result of bacterial flagellum under different conditions, such as the concentration and handling time of the suspension, staining time and staining methods, was compared. Results A slide of bacterial suspension with a concentration of 1.5- 10^12/L , to be placed at 37 ℃ for 10 min then stained by earbolfuehsin for 10-15 rain. Under a microscope, a clear background with red thalli and faint red flagellum could be seen in 80%--90% of the bacteria. Conclusion The concentration and handing time of suspension, slide-making method and staining time are important factors for an ideal flagellum staining.
