阿米卡星鞘内注射对中枢神经系统毒性影响的动物实验研究 被引量:4

摘要 目的探讨不同剂量阿米卡星单次鞘注对CNS的毒性影响。方法选用标准实验用兔24只,设生理盐水对照组,阿米卡星15mg,30mg,50mg实验组。分别在鞘注前后观察记录动物行为学变化、脊髓诱发电位(SCEP)和脊髓标本病理学检查。结果生理盐水组在鞘注前后各项观察指标差异无显著性。阿米卡星实验组全瘫率30mg组为33.3%,50mg组为66.6%,两组相比差异有显著性(P<0.01)。SCEP测定有不同程度潜伏期延长、波幅下降。实验组阿米卡星剂量和神经组织损伤程度间呈正相关(r=0.6746,P<0.05)。结论阿米卡星鞘注对兔CNS的毒性作用和剂量有相关性。 [Objective] To investigate the CNS toxic effect with different doses of intrathecal Amikacin. [Methods] 24 normal rabbits were randomly divided into 4 groups. 0.5 mL of diluent physiological saline and 15 mg, 30 mg, 50 mg Amikaein were injected into subaraehnoid seperately. The toxic effects were observed by ethology, spinal cord evoked potential (SCEP) and pathologic changes before and 'after intratheeal injection. [Results] No significant changes were observed of control animals with saline injected intrathecally. There was significant change of complete-paralyze rate between group C (33.3%) and group D(66.6%) (P 〈0.01). Amplitude of wave was cut down accompanied with the prolonged SCEP's PL in various degrees of treated rabbits. There was a positive relativity (r=- 0.6746, P 〈0.05) between doses of Amikaein and neuronal damages. [Conclution] The doses of intrathecal amikacin have a positive relativity with neurotoxic effeets of rabbits.
