脑出血灶周组织Bax、Bcl-2的表达与神经元凋亡关系的实验研究 被引量:13

摘要 目的检测脑出血灶周组织Bax、Bcl-2的表达,探讨与神经元凋亡的关系。方法对健康家犬30只在DSA介入下刺破大脑中动脉制作脑出血模型,选择出血后6h、12h、24h、48h、72h及96h共6个时间点,每点5只,均于相应时间点处死后取脑,检测出血灶周Bax、Bcl-2及神经元凋亡的表达。结果Bax及神经元凋亡的表达均于48h达高峰,至96h皆明显下降(与48h比较,P<0.01);Bcl-2出血后72h达高峰,至96h末见明显下降(与72h比较,P>0.05);Bax的表达与神经元凋亡呈正相关(P<0.05);Bcl-2与神经元凋亡的表达出血后48h前均呈正相关(P<0.05),48h后均呈负相关倾向,但无统计学差异(P>0.05)。结论Bax及Bcl-2参与了ICH后出血灶周神经元凋亡的过程,前者诱导和加重了神经元凋亡,后者有抑制凋亡作用,但显示其强度不足。 Objective To detect expression of Box and Bel- 2 in pefifocal tissue in cerebral hemorrhage and to study relationship between it and neuron apoptosis. Methods 30 healthy domestic dogs were stabbed in middle cerebral artery with DSA intervening, thus cerebral hemorrhage model was prepared. 6h, 12h, 24h, 48h, 72h and 96h after hemorrhage,6 time points altogether, were chosen. Each point included 5 dogs , which were executed at corresponding time point and then brain was taken out to detect expression of perifocal Box and Bcl - 2 and neuron apoptosis. Results Both expressions of Box and neuron apoptosis reached peak at 48h and there was marked decreasing at 96h (compared with that at 48h, both P 〈 0.01) .After hemorrhage,Bcl - 2 reached peak at 72h and there was no nmrked decreasing at 96h (compared with that at 72h, P 〉 0.05) .There was positive correlation between expression of Bax and neuron apoptosis( P 〈 0.05 ). Expressions of Bax and Bel - 2 and expressions of Bel - 2 and neuron apoptosis 48h after hemorrhage were positively correlated in each other (all P 〈 0.05 )and there was negative tendency after 48h. Yet there was no statistical differences (all P 〈 0.05). Conclusion Both Bax and Bcl - 2 make an important role in neuron apoptosis in perifocal tissue after ICH, and Box promotes or introdnees neuron apoptons, yet the latter one inhibits the expression of it.
