摘要
用高效毛细管电泳法分离环丙沙星、洛美沙星、氧氟沙星、氟罗沙星、帕珠沙星等5种喹诺酮类药物,探讨了缓冲液的浓度、pH值、分离电压、进样量对分离及检测的影响.实验发现采用50μm×40cm毛细管柱、5kV分离电压,用40mmol·L^-1Na2B1O7-KH2PO4缓冲液调至pH8.86时,上述5种组分可以在基线分离.在进样时间为5s、紫外检测波长为254nm的条件下,待测物的检测限介于1.5~7mg·L^-1之间,利用上述操作参数,采用标准加入法测定市售奥复星药片中氧氟沙星的质量分数为48.0%。
The aim of this work is to separate quinolones, namely, Lomefloxacin, Ciprofloxacin, Fleroxacin, Ofloxacin and Pazufloxacin, by means of high performance capillary electrophoresis. The parameters such as separation voltage, buffer concentration and pH, and injection time are studied. The separation conditions are.. 40 mmol·L^-1 sodium borate-potassium dihydrogen phosphate buffer at pH 8. 86, 50 gm×40cm(24cm effective length)capillary and 5 s injection; the separation voltage is+5 kV and the detection wavelength is set at 254 nm. The detection limits of the 5 quinolones are in the range of 1.5~7 mg·L^-1. Under the above experimental conditions, the effective content of the commercial Fleroxacin pill is detected to be 48.0%.
