β-巯基乙醇对培养胎儿卵巢组织异种移植效果的研究 被引量:3

摘要 目的探讨β-巯基乙醇(β-ME)对体外培养的卵巢组织的保护作用,寻找提高胎儿卵巢组织异种移植存活率的有效方法。方法取中晚期引产的死亡女胎,分别进行新鲜移植(对照组)或将卵巢组织在不含β-ME(培养对照组,β-ME0)、含100μMβ-ME(培养组,β-ME100)的培养液中新鲜培养3d后移植到裸鼠体内,从动情周期的恢复情况、组织学变化等方面比较各组移植物的活力。结果(1)β-ME0、β-ME100及对照组裸鼠动情周期的恢复率分别是62.5%、87.5%和33.3%,β-ME100组与对照组比较差异有统计学意义(P<0.05)。β-ME0、β-ME100组和对照组移植后动情周期开始天数为(25.20±4.49)d、(23.5±5.79)d和(12.75±1.5)d,两培养组均明显长于对照组(P<0.05);(2)异种移植后的组织学观察可见到存活移植物内有不同发育阶段的卵泡;卵泡计数β-ME100组(16.4±3.7815个/高倍视野)与对照组(4.0±2.8000个/高倍视野)比较差异有统计学意义,P<0.05。结论胎儿卵巢组织异种移植后卵泡可继续发育,经培养可提高移植物的活力,β-ME有利于提高移植物的存活率,促进卵泡的发育。 Objective To investigate the protective funtion of [β-Mercaptoethanol (β- ME) in the development of cultured and xenografted fetal ovaries, and try to find an efficient way to improve the suvival rate of fetal ovaries. Methods Fetal ovaries from the induced labor parturient in the second and third trimester of pregnancy were transplanted freshly (control group) or cultured with β- ME (100μMβ- ME,β- ME100 ) or without β- ME (0/1 Mβ- ME,β-ME0,experimental control group) for 3 days, then xenotransplanted the ovaries into the renal subcapsule of the recipients. The viability of xenografts was compared by observing the recovery of oestrus cycle and histological changes. Results (1) The recovery ratio of oestms cycle in β- MEO,β- ME100 group had a higher level (62.5%, 87.5%) than that of in freshly transplanted group (33.3%), P 〈 0.05, when compared with β- ME100 group (87.5%). The beginning days of oestrous cycle recovery in β- ME0, β- ME100 groups were (25.20±4.49,23.5 ± 5.79) longer than those of in control group (12.75± 1.5), P 〈 0.05. (2)The histological observation of xenografts found follicles in different periods in the survival grafts. The number of follicles in per high power field in two cultured groups were ( 12.1 ± 6.5345) and ( 16.4 ±3.781 ) respectively, compared with control group ( 16.4 ± 3.781 vs 54.0 ± 2.8), P 〈 0.05. Conclusion Fetal ovary tissue can develop after xenografting. The viability of fetal ovary tissue can be improved by means of culturing. β- ME can improve the survival ratio of grafts and follicles development.
