摘要 目的:研究胰岛素抵抗对犬血小板及红细胞功能的影响。方法:选12只Beagle犬,6只犬经高脂喂养6个月复制胰岛素抵抗模型,另6只作为对照,给予普通喂养,观察喂养前后生化指标、血小板及红细胞功能的变化。结果:高脂喂养6个月后,实验犬出现明显的代谢异常,即肥胖、胆固醇及甘油三酯明显升高(P<0.01)、氧化应激状态和胰岛素抵抗,并出现血小板过度活化的表现,即血小板聚集率明显增高(P<0.05),血小板表面黏附分子(P-Selectin和GPⅡb/Ⅲa)表达增加(P<0.01),血小板内钙离子浓度增加(P<0.05,P<0.01);同时,犬红细胞变形能力明显下降(P<0.05,P<0.01)。结论:胰岛素抵抗可造成犬血小板过度活化以及红细胞变形能力下降,代谢异常和氧化应激状态是导致血小板和红细胞功能异常的主要原因。改善血液流变学指标是胰岛素抵抗条件下血管保护的重要内容。 Objective :To investigate the impact of insulin resistance (IR) on functions of canine platelet and erythrocyte. Methods :6 Beagle dogs were fed with high-fat diet for 6 months to establish IR model; another 6 dogs were on routine diet and serving as control. Biochemical parameters and functions of platelet and erythrocytc were observed before and after the high-fat diet feeding. Results:After high fat diet feeding for 6 months, the dogs developed serious of metabolic disorders including obesity, dyslipidemia ( P〈0.01 ) , oxidative stress and insulin resistance. A platelet hyperactivity state was found characterized by increased aggregation and increased expressions of platelet surface adhesive molecules ( P-Selectin and GP Ⅱ b/Ⅲa) ( P〈0.01 ) ; platelet intracellular calcium concentration elevated ( P〈0.01 or P〈0. 05 ) ; erythrocyte deformability was decreased remarkably(P〈0.05 or P〈0.01 ). Conclusions:Insulin resistance would result in platelet hyperactivity and decreased erythrocyte deformability. These might be caused mainly by metabolic disorder and oxidative stress. Therefore,one of the important steps to protect vessels was to improve hemorheology under IR.
