摘要
目的观察尼美舒利、舒林酸对人胃癌SGC-7901细胞环氧化酶2(COX-2)和血管内皮生长因子(VEGF)表达的影响,初步探讨尼美舒利、舒林酸抑制肿瘤血管生成机制。方法对人胃癌SGC-7901细胞进行细胞培养,免疫组织化学、蛋白质印迹(Western blot)方法检测不同浓度尼美舒利、舒林酸作用后细胞COX-2、VEGF表达的改变。结果尼美舒利100、200μmol/L及舒林酸0.6、1.2 mmol/L作用48 h后人胃癌SGC-7901细胞COX-2、VEGF蛋白表达率与阴性对照组相比明显降低(P<0.05),并且COX-2与VEGF之间具有相关性(P<0.05)。随着药物浓度的升高COX-2和VEGF表达逐渐减弱。结论抑制COX-2的表达及由此引起的VEGF蛋白表达降低,在非甾体类抗炎药(NSAIDs)抑制肿瘤血管生长机制中可能具有一定作用。
Objective To explore the effect and significance of nimesulide and sulindac on the expression of COX2 and VEGF. Methods SGC-7901 cells were treated with different concentrations of nimesulide and sulindac for 48 hours, then the expressions of COX-2 and VEGF in tumor cells were detected with Power VisionTM 2 step immunohistochemical method and Western blot analysis. Results The expression rates of COX-2 and VEGF in 100 and 200μmol/L nimesulide and 0. 6 and 1.2 mmol/ L sulindac group were significantly decreased more in gastric cancer group than those in control group (P〈0. 05). The expression of COX-2 was positively correlaed with that of VEGF. The expressions of COX-2 and VEGF were decreased as the drug concentions went down Conclusion The inhibition of the expression of COX-2 and VEGF may play a certain role in the antitumor process of NSAIDs.
