The application of dynamic implantation and culture in vascular tissue engineering

摘要 目的研究体外构建组织工程血管的方法。方法以兔的血管内皮细胞和平滑肌细胞为种子细胞,在聚β羟基丁酯构成的血管支架上先后种植,比较静态和动态种植、静态和动态培养两种条件下细胞种植吸附率,分布及细胞增殖的差异。结果运用旋转成管法,成功制备了血管支架;培养的第2、4周动态组细胞增殖较静态组快,细胞排列有序,顺培养液流动的方向排列,而静态组细胞增殖慢,排列不规则。结论动态种植优于静态种植,动态培养优于静态培养。 Objective To study the method for vascular tissue engineering in vitro. Methods Vascular endothelial ceils and smooth muscle ceils of rabbits were cultured, then implanted in scaffolds of PHB sequentially. Static and dynamic implantation and culture method were compared. Results Tubular vascular scaffolds were successfully made by rotation method. The cells grew faster in dynamic group than that in static group. The cells arranged regularly in culture liquid flowing direction in dynamic group, but irregularly in static group. Conclusion Dynamic implantation and culture method is superior to static one.
