摘要
目的观察生长抑素类似物奥曲肽在体内对胃癌生长的影响。方法人胃癌细胞株SGC7901细胞接种于48只裸鼠背部皮下制成荷瘤动物模型。第8天将裸鼠随机分成4组:奥曲肽(Oct组,100μg/kg;s.c.qd);5Fu组(17mg/kg,ip,2次/周);奥曲肽和5Fu联合治疗组(联合治疗组);对照组,每组各12只裸鼠。连续用药6周。末次用药24小时后处死动物,检测肿瘤体积、重量、坏死体积,比较抑瘤率。结果奥曲肽组、5Fu组、联合治疗组、对照组的平均瘤体体积(cm3)分别为2.17±0.78、2.19±0.79、1.36±0.75、3.23±0.74(F=9.317,P=0.0001);平均瘤体重量(g)分别为6.88±2.06、7.22±2.47、4.47±2.28、10.30±2.80(F=5.452,P=0.0001);平均坏死体积(cm3)分别为0.55±0.38、0.52±0.53、1.14±0.83、0.32±0.27(F=13.987,P=0.0001);奥曲肽组、5Fu组、联合治疗组的抑瘤率分别为:33.20%、29.92%、56.60%(χ2=6.461,P=0.04)。结论奥曲肽可以抑制裸鼠胃癌移植瘤的生长。
Objective To investigate the effcets of somatostatin analogue octreotide on the growth of gastric cancel SGC-7910 in vivo. Methods The forty-eight nude mice of xenotransplanted human gastric cancer cell line SGC-7901 in gymnomouse body were set up. On the eighth day, all nude mice were randomly divided into four groups and each group had twelve mice. Octreotide at a dose of 100ug/kg was subcutaneously administered once a day in Octreotide group. 5-FU was intraperitoneally administered twice a week in 5-FU group. Combined group receivel all the reagents as Octreotide group and 5- FU group. Control group received injection of NS. Six weeks later, all mice were executed. The tumor mass volume, weight, necrosis volume were observed and the inhibition rate of tumor growth were also caculated, Results The average tumor vol- umes(era3) in Octreotide group, 5-FU group, combined group and control group was 2.17 ± 0.78,2.19 ± 0.79,1.36 ± 0.72 and 3.23 ±0.74 respectively ( F = 9.317, P = 0.0001) .The average tumor weight (g) was 6.88 ± 2.06, 7.22 ±2.47, 4.47 ± 2.28 and 10.30 ± 2.80( F = 5.452, P = 0.0001 ). The average tumor necrosis volume(era3) was 0.55 ± 0.38, 0.52 ± 0.53, 1.14 ± 0.83 and 0.32 ± 0.27( F = 13.987, P = 0.0001 ), and the average tumor inhibition rate in Octreotide, 5- FU and Combined group was 33.20% ,29.92% ,56.60% respectively( X^2 = 6.461, P = 0.04). Conclusion Octreotion could inhibit the SGC-7901 growth in vivo.
