变应性鼻炎动物模型白细胞介素12与嗜酸性粒细胞的变化 被引量:9

摘要 目的本文观察白细胞介素1(2interleukin-12,IL-12)在变应性鼻炎动物模型中的变化及其与嗜酸性粒细胞的相关性。方法39只6~8周雄性BALB/c小鼠,随机分成自然对照组、变应性鼻炎(allergicrhinitis,AR)组和布地奈德治疗组,用卵清蛋白致敏激发制备实验性变应性鼻炎动物模型,取鼻黏膜制备石蜡切片后进行HE常规染色和IL-12免疫组化染色并显微镜观察计数。结果AR组嗜酸性粒细胞计数极显著性地大于自然对照组,P<0.01。布地奈德治疗后组织中嗜酸性粒细胞少见,极显著性地低于AR组,P<0.01;而IL-12的表达显著性地高于AR组,P<0.05,但与自然对照组相比无明显差别,P>0.05;组织中IL-12的表达与嗜酸性粒细胞浸润有一定的负相关性,R=0.9252。结论鼻黏膜组织中IL-12的异常表达,在变应性鼻炎的发病机制中起重要作用,恢复或提高局部组织中IL-12的表达,能抑制嗜酸性粒细胞在鼻黏膜中的浸润,改善变应性疾病的症状。 OBJECTIVE To observe the expression of interleukin-12 and eosinophils in the nasal mucosa of allergic rhinitis mice. METHODS Thirty nine male BALB/c mice, 6-8 weeks old, were randomly divided into three groups: control group, allergic rhinitis (AR) group, and Budesonide treatment group. Allergic rhinitis model in mice were established by using ovalbumin intraperitoneal immunization and nasal antigen challenge. The nasal mucosa obtained from mice of three groups were stained routinely by HE and immunohistochemical method to observe the distribution and expression of intedeukin-12 and eosinophils. RESULTS The expression of eosinophils in the nasal mucosa of AR group was significantly higher than control group (P〈0.01). After treatment with Budesonide, eosinophils' expression were decreased in nasal mucosa (P〈0.01, compared with the AR group). However, interleukin-12 expression was significantly increased compared with AR group (P 〈0.01), and had no significant difference compared with the control group. There was a negative relationship between the expression of interleukin-12 and eosinophil in nasal mucosa (R =0.9252). CONCLUSION The abnormal expression of interleukin-12 in the nasal mucosa played an important role in the pathogenic mechanism of AR. The recovery or increase expression of interleukin-12 in topical tissue in vivo could suppress the expression of eosinophils and may improve the therapeutic effectiveness of AR.
