人星形细胞瘤细胞系JCBI的建立及其特性观察

ESTABLISHMENT OF A HUMAN ASTROCYTOMA CELL LINE (JCBI) AND INVESTIGATION OF ITS CHARACTERISTICS

一株人类星形细胞瘤细胞系在体外已连续传代培养近3年,传至180代。定名为JCBI。其形态具有恶性肿瘤细胞的典型特征;细胞倍增时间为70h;培养第6天时细胞分裂指数为33‰;平板集落形成率为49％;染色体数是83条(35％)和82条(27％),为亚四倍体核型,染色体具有明显异常;在免疫抑制小鼠皮下异种移植成功,成瘤率为87.5％;电镜观察JCBI细胞浆内含有丰富的胶质微丝。用抗GFAP抗体进行免疫细胞化学染色,发现JCBI细胞呈GFAP阳性,证明JCBI确为星形细胞来源的恶性肿瘤。本细胞系的建立为胶质瘤研究提供了一个较好的细胞实验模型。

A new continuous cell line arising from an astrocytoma (grade Ⅱ°)was established. It has been subcultured for 180 generations in a period of 3 years. This cell line was designated as JCBI. Morphologically this line showed the properties of tumor cells. In vitro, the doubling time was 70 hours, and the mitotic indexwas 33‰ on the 6th day. The mean plating efficiency was 49%. The number of chromosomes varied in different cells, from 73 to 100, but the majority of cells contained 83 (35%)or 82 (27%) chromosomes, a lot of which were abnormal.The cultured cells were readily tumorigenic in mice. Plenty of glial fibrillary filaments were seen under electron microscope. It was proved that JCBI cells were GFAP positive by immunocytochemical analysis with anti-GFAP serum.
