瘤内注射复方中药99-克星诱导肝癌细胞凋亡的体外实验研究

Ultrasound-guided percutaneous intratumoral injection of Chinese medicine “Star-99” in the apoptosis of human liver cancer cell line HepG_2: an experimental study

目的探讨复方中药99-克星(99-克星)直接诱导人肝癌细胞系HepG2凋亡的作用。方法应用透射电镜、原位末端标记技术(TUNEL)等方法,观察99-克星直接作用于HepG2细胞后的形态学改变。结果99-克星直接作用于HepG248h后肝癌细胞出现典型的凋亡征象,可分别从光学显微镜(HE染色)及透射电镜下观察到肿瘤细胞凋亡现象,凋亡细胞表现为单个或散在分布的肿瘤细胞胞浆呈较强嗜伊红色,核致密浓缩出现核碎裂或核染色质断裂而形成大小不等的凋亡小体,TUNEL法检测到发生在各期的凋亡细胞。结论99-克星可直接诱导HepG2细胞凋亡,可作为一种新的细胞凋亡诱导剂用于肝癌的治疗,有广阔的应用前景与价值。

Objective To study the role of Chinese medicine ＂Star-99＂ in direct induction of the apoptosis of human liver cancer cell line HepG2. Methods Using HE staining, transmission electron microscopy （TEM） and TdT-mediated dUTP-biotin nick end labeling （TUNEL） methods, the morphological changes of the apoptosis of HepG2 cells induced directly by Star-99 were detected. Results After direct exposure to ＂Star-99＂ for 48 h, the live cancer cells showed typically morphological and biochemical features of apoptosis under light microscope （ by HE staining） and TEM. The liver cancer cells became round and shrunk, then the cells fell to pieces and its refractions floated up, the cytoplasts became eosinophilous and the karyons condensed and became bluish dark by HE staining. The karyons fragmented or the karyotin broke down to form apoptotic corpuscles of varying sizes. Apoptotic cells in different phases were observed by TUNEL. Conclusions Chinese medicine ＂Star-99＂ can directly induce extensive apoptosis of HepG2 cells. The apoptotic effect of ＂Star-99＂ on tumor cells may play a key role in its anticancer activities, thus ＂Star- 99＂ is a new and promising apoptosis-inducer in the treatment of liver cancer.
