摘要
用孕马血清(PMSG)和人绒毛膜促性腺激素(hCG)对6—8周龄的雌性昆明系及ICR系小鼠进行超数排卯处理,从注射hCG后10h起每间隔2h采集小鼠的卵丘卵母细胞复合体(COCs),对卵母细胞的第一极体(PbI)进行数量和形态分级统计,并用台盼蓝染色法鉴定第一极怵的活力;同时,对新获取的卵丘卵母细胞复合体进行不同温度保存条件试验。结果表明,在注射hCG后12h所获得的第一极体活性最高;在室温和37℃下,第一极体的活力在6h后便完全丧失,而在4℃下保存的第一极体,经过48h后其活力依然良好。
Pregnant mare serum gonadotrophin (PMSG) and human chorionic gonadotrophin(hCG) were used to stimulate super ovulation of Kunming and ICR mice of 6-8 weeks old. Ten hours after injection of hCG, cumulus-ooeyte complexes(COCs) were collected from each group of mice every 2 hours. The number of the first polar body(PbI) were checked, and morphological classes of PbI were counted. The vigor of PbI was observed following Trypan blue staining. Surviving time of PbI at different temperature was investigated. The result showed that 12 hours after hCG treatment the PbI had the best vigor of all groups. Phi died after 6 h at 37℃ or at room temperature and could survive more than 48 h at 4℃.
