吸入一氧化氮对胎粪吸入性肺炎肺组织Ⅰ、Ⅲ型胶原mRNA表达的影响 被引量:1

Effects of iNO on mRNA Expression of Lung Collagen Ⅰ and Ⅲ of Meconium Aspirated Rabbits

摘要 目的 探讨吸入一氧化氮对胎粪吸人性肺炎肺组织Ⅰ、Ⅲ型胶原mRNA表达的影响。方法 通过建立实验性胎粪吸人性肺炎家兔模型,在不同氧浓度下以不同浓度的NO吸入干预12h后,应用RT-PCR方法测定肺组织Ⅰ、Ⅲ型胶原mRNA表达。结果 各组之间Ⅰ、Ⅲ型胶原mRNA表达水平均无显著差别(P〉0.05),各组Ⅰ型胶原/Ⅲ型胶原比值也无差异(P〉0.05)。结论 短期(12h)内不同氧浓度下6~20ppmiNO干预对肺组织Ⅰ型胶原、ⅢmRNA表达尚无影响。 Objective To discuss the effects of iNO on the number of white blood cells and level of IL-8 in the BALF of meconium aspiration pneumonia animals. Methods Animals were randomly allocated to 10 groups (n = 60) of 6 each. Control or meconium aspiration pneumonia model groups were inhaled with room air or 100% O2.6 treatment groups were treated with continuous NO inhalation at a dose of 6,10 or 20 × 10^-6 respectively for 12 hours under room air or 100% 02. Expressions of lung collagen Ⅰ and Ⅲ mRNAs were analyzed by RT-PCR. Results Expressions of lung collagen Ⅰ and Ⅲ mRNAs and ratio of collagen Ⅰ/collagen Ⅲ showed no significant difference between the studied groups(P〉0.05 respectively ). Conclusion In summary,there is no effect on the expressions of lung Ⅰ and Ⅲ collagen after inhaled NO for 12 hours, which may be due to short time exposure to iNO under 21% 02 or 100%O2.
