摘要
目的探询脑室定向注射淀粉样蛋白对大鼠大脑皮层神经元驱动蛋白超家族(KIFs)基因表达的影响。方法脑室定向注射淀粉样蛋白诱导大鼠产生类似阿尔茨海默病的症状。水迷宫实验验证大鼠记忆损伤程度;胆碱乙酰基转移酶(ChAT)基因的RT-PCR扩增证实大鼠大脑皮层胆碱能神经元损伤。实时荧光PCR方法检测大鼠大脑皮层神经元驱动蛋白(kinesin)、KIFlb、KIFlc、KIF3a、KIF3c与KIF4基因的表达。结果水迷宫实验与ChAT表达检测表明动物模型制备成功。实时荧光PCR结果显示,淀粉样蛋白脑室内注射造成大鼠皮层KIFs基因表达的普遍上升。结论淀粉样蛋白造成的皮层胆碱能神经元损伤很可能与其兴奋性毒性有关。
Objective To investigate the effect of amyloid on the expression of kinesin superfamily(KIFs) genes in cortex of Wistar rats. Methods The animal model of Alzheimer' s disease was prepared by injection of amyloid into brain ventricle of rats. The learning impairments were verified by water maze test. The levels of mRNA for six kinesin superfamily genes, kinesin, KIF1b, KIF1c, KIF3a, KIF3c, and KIF4, in cortex of rats, were estimated by real-time RT-PCR. Results Two weeks after amyloid injection, the rats showed significant learning impairments. In cortex, the expression of KIF genes was increased after amyloid injection. Conclusion The increase of KIF gene may be an evidence for the excitotoxity of amyloid to neurons.
