异种脱细胞真皮修补巩膜缺损实验研究

摘要 目的观察异种(猪)脱细胞真皮(Xeno-ADM)修补兔眼巩膜缺损的临床效果及病理组织学变化。方法制作兔局部巩膜缺损模型,用Xeno-ADM进行修补,活体观察动物术后情况及移植物情况,并分别于第2、4周取移植交界处移植物光镜下观察炎症变化、血管化情况及融合情况。结果大体观察未见明显炎症反应及眼球的变形溶解;光镜下2周时可见局部炎细胞浸润,Xe-no-ADM与自体巩膜形成局部融合,4周时炎细胞明显减少,Xeno-ADM与自体巩膜完全融合。结论异种脱细胞真皮血管化快、免疫原性低,是一种良好的巩膜替代物。 Objective To observe clinical results and histopathologieal changes of scleral defect repaired with acellular xenogenic dermal matrix (Xeno-ADM) in rabbits. Methods Model of rabbit sclera defect was established, and repaired with Xeno-ADM. The rabbits were clinically examined for inflammation and eyeball healing. The animals were sacrificed at 2nd and 4th week after operation, and implants were obtained and examined histopathologyically and ultrastructully with light microscopies respectively to evalua ting changes of inflammation and vascularization. Results There were no obvious inflammation and eyeball deformation observed. 2 weeks after implantation, the partial inflammatory cell invasion was seen with the light microscopy, and there was an obvious border line between the Xeno-ADM and the selera. 4 weeks after implantation, the inflammatory cells were reduced noticeably, the Xeno ADM and the selera completely merged with each other. Conclusion The aeellular xenogenic dermal matrix may be an ideal materix with fast neovascularization and low immunity for replace of sclera implants.
