人参糖蛋白的分离纯化及其性质研究 被引量:4

摘要 目的:分离纯化人参糖蛋白组分 LRC 并研究其理化性质及初步的结构特性。方法:采用 HiPrep 2660 Sephacryl S200和Hitrap Q^(TM) 26/60柱色谱得到均一的糖蛋白组分,高压柱层析法、凝胶电泳法及3种分离原理的毛细管电泳法检测纯度,薄层色谱和气相色谱分析单糖组成,红外光谱确定糖苷键类型。结果:LRC 为均一组分,单糖组成为鼠李糖、阿拉伯糖、葡萄糖和半乳糖,其摩尔比为1.00:3.45:0.612:5.03,总糖含量为32%,蛋白含量为60%,由门冬氨酸、谷氨酸等17种氨基酸组成,糖肽键为苏氨酸连接的 O-型糖肽键。结论:LRC 是以α-吡喃糖为主的糖蛋白,相对分子质量为2.4×10~4。 Objective: To purify a glycoprotein named LRC from Panax ginseng and to investigate its properties and its elementary structure. Methods:LRC was purified by hiPrep 2660 Sephacryl S200 and Hitrap QTM 26/60 column chromatography. The homogeneity of LRC was checked by CZE, HPGPC, CGE, CIEF and PAGE. Sugar composition was determined by TLC and GC. The saccharide chain was concluded by IR. Results:LRC was homogeneous and consisted of Rha,Ara,Glc and Gal in the molar ratio of 1.00: 3.45: 0. 612: 5.03. Its saccharide and protein were 32% and 60% respectively. Its protein portion consisted of Asx ,Glx and other 15 kinds of amino acids. The saccharide and protein of LRC was linked by O - glycosidic bond. Conclusion:The characteristic absorption in IR spectrum is indicative of the α - D - pyranan linkage. Its relative molecular mass is about 2. 4 × 10^4.
