摘要
目的研究短期吸入二氧化硫(SO2)毒理作用的分子机制。方法采用Affymetrix基因芯片(RAE230A)技术,研究SO2短期(56 mg/m3,6 h/d,共7 d)吸入后大鼠肺组织基因表达谱的改变。结果与其相应的对照组相比,SO2短期吸入后表达上调的基因有31个,其中包括18个已知基因和13个新基因,而表达下调的基因有30个,其中包括19个已知基因和11个新基因。结论(1)短期吸入SO2的大鼠肺基因表达谱与其对照组相比,氧化磷酸化相关的一些基因发生了变化,表明高剂量短期吸入SO2可能导致线粒体功能损伤;(2)与甲状腺激素有关的几个基因表达也发生了变化,表明SO2可能会造成甲状腺激素在体内生理过程的改变;(3)SO2可引起多种基因的表达发生改变,这意味着多种基因的表达是易变且不稳定的,在肺组织中可能存在着SO2诱发的表达不稳定型基因组。
Objective To study the molecular mechanism of toxicological effects of short-term exposed to sulfur dioxide (SO2)on mammalian animals and humans. Methods Affymetrix Genechip(RAE230A) analysis was carried to analyze the gene expression profiles in lungs of Wistar rats exposed to SO2 for short-term(56 mg/m^3, 6h/day, for 7 days), Results It was found that 31 genes, containing 18 known genes and 13 novel genes, were up-regulated, and 30 genes, containing 19 known genes and 11 novel genes, were down-regulated in rats short-term exposed to SO2 compared with control rats. Conclusion (1)A notable feature of the gene expression profile was the changed expression of genes related to oxidative phosphorylation in lungs of rats short-term exposed to SO2, which showed high-dose short-term exposed to SO2 may cause the deterioration of mitochondrial functions. (2)Expressions of genes related to thyroid functions in lungs of rats were also changed by high-dose short-term exposed to SO2. (3)Expressions of many genes in rat lung were changed by SO2, it implied that the gene expressions were changeful and unstable, and there was a genome that its expression might be changed by SO2.
