异甘草酸镁对培养肝细胞增殖影响的实验研究 被引量:81

摘要 目的观察异甘草酸镁对肝细胞增殖的影响。方法用含不同浓度异甘草酸镁的培养基(0、0.001、0.01、0.1、1.0、10.0、100.0mg/ml)培养肝细胞,通过绘制细胞生长曲线和四甲基偶氮唑盐法(MTT法)检测异甘草酸镁对肝细胞增殖的影响,并同时观察其形态变化。结果在所设加样量范围内,与空白对照组(0mg/ml)比较,培养基所含的异甘草酸镁浓度为1.0mg/ml时,对张氏肝细胞具有明显的促进增殖作用;10.0mg/ml时,其促增殖作用达到最高峰,而100.0mg/ml时,则出现明显的抑制作用。结论异甘草酸镁对肝细胞的增殖具有明显的影响,在一定剂量范围内对肝细胞具有明显的促进增殖作用。 Objective To investigate the effects of magnesium isoglycyrrhizinate on the proliferation of hepatocytes. Methods Hepatocytes were incubated with various concentrations of magnesium isoglycyrrhizinate solution(0 mg/ml, 0.001 mg/ml, 0.01 mg/ml, 0.1 mg/ml, 1 mg/ml, 10 mg/ml, and 100 mg/ml). With the cell growth curve and 3-(4, 5-dimethylthiazol-2-yl)-2, 5- diphennylterazolium bromide (MTT) colorimetrlc assay the effects of maganesium isoglycyrrhizinate on the proliferation of cnltured hepatocyte were studied, and morphological changes of the hepatocytes were also observed. Results Compared with the control group(0 mg/ml group) ,the proliferation of hepatocytes significantly increased when the concentration of magnesium isoglycyrrhizinate in medium reached 1.0 mg/ml. The proliferation of hepatocytes presented its peak when the magnesium isoglycyn'hizinate concentration was 10.0 mg/mI. But, the proliferation of hepatocytes was significantly inhibited when the concentration of magnesium isoglycyrrhizinate in medium reached 100.0 mg/ml. Conclusions Magnesium isoglycyrrhizinatc could significantly promote the proliferation of bepatocytes within certain concentration range.
