黄孢原毛平革菌产生的具有氧化性活力低分子肽类物质的分离纯化与性质鉴定 被引量:7

摘要 从黄孢原毛平革菌(Phanerochetechrysosporium)发酵液中分离纯化到一个新的具有酚氧化活力的低分子组分,命名为Pc因子.该组分分子量在600Da左右,含Glu,Gly,Val三种氨基酸残基,其紫外和红外光谱也显示Pc因子含有氨基酸,氨基酸的Cα信号在13C-NMR方法中也可检测到.该物质对热稳定,在酸性条件下易失活,弱碱性条件下保持稳定.它具有螯合Fe3+的能力,并能够将Fe3+还原为Fe2+,但不能产生羟基自由基HO·.在不需要Mn2+和H2O2激活的条件下能够氧化酚型木素模型物2,6-二甲氧基酚(2,6-dimethoxyphenol,2,6-DMP),2,2′-连氮二(3-乙基苯并噻唑-6-磺酸)(2,2′-azinobis(3-ethylbenzathiazoline-6-sulfinicacid),ABTS)和丁香醛连氮,其氧化作用需要金属离子和O2的参与,为木素生物降解过程中的一类新的氧化机制,Pc因子在此氧化还原体系中执行电子传递体的功能.
