摘要
目的:建立兔耳痤疮动物实验模型,用中药制剂消痤霜外涂,观察其疗效。方法:①实验于2003-07/08在北华大学医学院实验动物中心实验室完成。选用成年家兔20只。②建立兔耳痤疮动物模型:于每只家兔左右耳内侧耳管开口处2cm×2cm范围,每日涂煤焦油1次,每次0.5mL,连续2周。③将20只家兔随机分为2组:实验组与对照组,各10只,实验组右耳痤疮模型用消痤霜擦剂(主要成分为大黄、黄连、金银花、龙葵、积雪草等,由本院中药制剂室制备成外涂水剂,呈淡黄色),2次/d,每次约1mL擦涂,连续2周。对照组涂抹生理盐水。④14d后对两组兔右耳及实验组兔左耳痤疮模型处皮肤做肉眼观察及病理活检,进行组织学观察。痤疮模型组织学判定分级标准:0级为无痤疮表现;1级为毛囊漏斗部可见少量致密角化物质;2级为毛囊漏斗部可见中等致密角化物质,并向皮脂腺延伸;3级为扩张的毛囊内有广泛的角化物质,人类痤疮相似。⑤计量资料差异比较采用t检验。结果:实验过程中对照组死亡1只,最终进入结果分析19只。①实验组涂抹消痤霜2周后,肉眼见:局部毛囊角栓减少,部分角栓脱落后遗留点状凹陷的毛囊,丘疹变平减少;组织学观察见:表皮增厚明显减轻,真皮炎症细胞明显减少,但仍见毛囊口轻度扩张,有少量疏松角化物质充填。②对照组实验2周后肉眼仍可见明显的毛囊角栓,但丘疹减轻或变平;组织学观察见:角化过度,真皮内仍见较多的炎症细胞浸润,毛囊口及漏斗部仍见角化物质堆积,漏斗部扩大如壶状。③实验组治疗后痤疮模型组织学级别明显低于对照组犤(0.6±0.52),(2.5±0.53)级,P<0.01犦。结论:中药制剂消痤霜对兔耳痤疮实验模型具有减少毛囊的角栓、减轻真皮炎症和抑制瘢痕形成的作用。
AIM: To observe the effect of xiaocuo shuang, a Chinese herb, on animal acne model of rabbit's ears. METHODS: ① The experiment was completed in the Experimental Animal Center of Medical College of Beihua University from July to August 2003. Twent3, adult rabbits were selected. ② Establishment of animal acne model of rabbit's ears: Pix carhonis was smeared at 2 cm × 2 cm opening site inside left and fight ear of rabbit with 0.5 mL once per day for 2 successive weeks. ③ Totally 20 rabbits were randomly divided into experimental group and control group with 10 in each group. Acne models of right ear in experimental group were smeared with xiaocuo shuang of 1 mL once and twice per day for 2 successive weeks, and models in control group were smeared with saline. Main components of xiaocuo shuang were rhubarb, coptis root, honeysuckle flower, nightshade and jixuecao, and xiaocuo shuang was made into smeared water in light yellow. ④After 14 days, skin of acne model of right ear in the two groups and left ear in experimental group was observed by naked eye, checked livingly, and observed with histological method. Histological classified criteria of acne model: 0: no acne; 1: a few of dense cornification was observed at infundibulum of hair follicle; 2: a little more dense comification was observed at infundibulum of hair follicle and extended to sebaceous gland; 3: a lot of dense cornification was observed in ectatic hair follicle and was similar to acne of human. ⑤ Differences of measurement data were compared with t test. RESULTS: Totally 19 rabbits entered the final analysis except 1 rabbit in control group died in the experiment. 03 Two weeks after smearing xiaocuo shuang in experimental group, it was observed by naked eye that spline in local hair follicle was decreased, hair follicle shaped in points and incisure was leaved after abscission of local spline, and papules were relived; with histology that thickness of cuticle was relieved, inflammatory cells of corium were reduced, but opening of hair follicle was expanded lightly, and a few rarefaction comification was filled in. ② Two weeks after smearing xiaocuo shuang in control group, it was observed by naked eye that spline of hair follicle was obvious, and papules were relived; with histology that hyperkeratosis was obvious, inflammatory cells of corium were still survived, stack of cornification was still found out at opening of hair follicle and infundibulum, and infundibulum was expanded grated and shaped into pail. ③ After treatment, histological grades of acne model were lower in experimental group than those in control group [(0.6±0.52), (2.5±0.53) grades, P 〈 0.01]. CONCLUSION: Xiaocuo shuang a Chinese herb can decrease spline of hair follicle, relive derma inflammation and inhibit formation of scar obviously on aene model of rabbit's ears.
