摘要
目的研究应用离子色谱法测定血浆和尿液中麦芽糖的条件,探讨该方法的性能及临床实用性。方法采用带脉冲电化学检测器的高压液相离子色谱仪,层析系统由GP40梯度泵和ED 40电化学检测器组成。测量电极为金电极,Ag-AgCl为参比电极。分离柱为CarboPac-PA1阴离子柱。流动相A为0.1mol/L NaOH,流动相B为0.5 mol/L AcNa/0.1 mol/L NaOH。结果血浆和尿样高、中、低3个浓度检测的变异系数分别为:血浆批内CV: 0.17-6.38(n=5):批间CV:3.68~4.68(n=25)。尿样批内CV:0.34-9.15(n=5);批间CV:2.91-7.63(n=25)。血浆回收率为97.41%-102.02%,尿样回收率为100.48%-107.06%。血浆和尿样标准曲线的线性范围分别是0.25-40μg/ml 和0.5-160μg/ml,最低检测线为0.125μg/ml。结论该方法灵敏度高,取血量少,可以作为临床研究和检验的方法。
Objective To explore the performance and clinical application of Ion-exchange chromatography in measuring the maltose levels in human plasma and urine. Methods Dionex 500 high pressure liquid ion chromatography was used. It was composed of GP 40 gradient pump and ED 40 electrochemical detector. The gold electrode was designed as a working electrode, and pH Ag/AgCl as reference electrode. Flowing phase A indicated 0.1 mol/L NaOH, and B was 0.5 mol/L AcNa /0.1 mol/L NaOH. Results The method had a good reproducibility. The intra-assay coefficients of variation (CVs) among low, middle, and high concentrations of maltose in plasma and urine were 0.033-6.28 and 0.34-9.15, respectively, while the inter-assay CVs ranged 3.68-4.68 and 2.91-7.63. The average recovery rates of plasma and urine were 97.41%-102.02% and 100.48%-107.06%, respectively. The lineal range were 0.25-40 μg/ml in plasma and 0.5-160μg/ml in urine. The lowest determination limit was 0.125μg/ml. Conclusion liquid ion chromatography is stable, simple, rapid, sensitive, and accurate in determining the in human plasma and urine and is suitable for researches and clinical application.
