矽肺纤维化模型大鼠肺组织基质金属蛋白酶-2和基质金属蛋白酶-9的动态表达 被引量:7

摘要 ①目的探讨矽肺纤维化模型大鼠肺组织MMP-2和MMP-9表达的变化。②方法清洁级W istar大鼠50只,随机分为对照组和模型组,每组25只。采用气管内注射S iO2悬液方法复制大鼠矽肺纤维化模型,对照组代以等剂量生理盐水。注射后1、3、7、14、28天取出肺组织,应用免疫组织化学和RT-PCR方法,观察其MMP-2、MMP-9蛋白及其mRNA的动态变化。③结果模型组肺组织MMP-2和MMP-9表达均较对照组增高。④结论S iO2粉尘可通过高表达MMP-2和MMP-9参与矽肺纤维化的发生和发展。 Objective To study the expression of MMP - 2 and MMP -9 in lung tissue of rats pulmonary.fibrosis model. Methods 50 Wistar rats were divided randomly into control group and model group. Model groups were instillated with SiO2 , while control group with normal asline instead. Lungs were isolated at day 1,3,7,14,28th after SiO2 exposure. Expression of MMP -2 and MMP- 9 were evaluated by Immunochemistry and RT- PCR. Results The MMP- 2 and MMP- 9 expression in lung tissue in model group increased higher than that of control group. Conclusions SiO2 might take part in the pulmonary fibrosis by effect the expression of MMP- 2 and MMP-9.
