氧化亚甲蓝-碘化钾法髓过氧化物酶快速染色及应用

摘要 目的:介绍一种安全稳定、快速可靠的髓过氧化物酶(MPO)染色新方法(氧化MB-KI法)。方法:采用亚甲蓝(Methylene blue)为显色剂取代Pereira原法中的Leishman's染料,并采用人工预先氧化亚甲蓝溶液的新方法,使其一步到位达到最佳氧化成熟程度,以后每次配制MPO染色工作液时不必加H2O2。结果:本法(氧化MB-KI法)MPO阳性产物呈棕黑色颗粒状,细胞核为蓝绿色。与Washburn联苯胺法比较,两者阳性率十分相近,差异无显著性意义(P>0.05)。结论:氧化MB-KI法所显示的MPO阳性反应对比鲜明、易于判断,避免了假阴性结果的发生。试剂安全、稳定可靠、易于配制、操作简单、染色时间短,是理想的MPO染色方法。 Objective:To introduce a new method of myeloperoxidase (MPO) staining characterized by safety, stability celerity and credibility. Method: Use Methylene blue instead of Leishman's of the original, and make it attain best oxidized degree in one step by oxidizing Methylene blue solution with artifical method in advance. There is no need to add H2O2 into MPO staining solution when making it. Result: The positive product of this method (oxidized MB-KI method) appears brown and black granules, and the nucleus are blue and green. The positive rate of this method was nearly as same as that of Washburn MPO, and there were no significant differences ( P 〉 0.05). Conclusion:The positive reaction of the oxidized MP-KI method is clearer, which is much easier to be iden tified and can avoid the false-negative results. The reagent is safe, steady and credible. It is easy to make and op erate, and takes shorter time to stain. It is an ideal method of MPO staining.
