利用DAS-ELISA检测侵染观赏百合黄瓜花叶病毒的研究 被引量:5

摘要 从制备的黄瓜花叶病毒———观赏百合分离物(CMV-Li)抗血清中提纯了IgG,并用过碘酸钠法进行辣根过氧化物酶(HRP)标记,建立了检测CMV-Li的DAS-ELISA(double antibody sandwich-enzyme-linked immumnosor-bent assay)系统,利用此检测系统并结合生物学方法,对临洮切花百合田间病样和百合不同部位带毒量进行了检测,结果发现CMV-Li的侵染率为23.81%,叶片的带毒量明显高于花和鳞茎. Immunoglobulin (IgG)from antiserum of ornamental lily that infected by cucumber raosaic virus was purified and marked with horseradish peroxidase (HRP) by sodium periodate. The method of DAS-ELISA for CMV-Li detection was developed. Some field samples of ornamental lily from Lintao and Yuzhong were detected by DAS-ELISA,the results showed that CMV-Li infected lily was a common pathogen of lily. About 23.85 % of detected lily samples both in Lintao and Yuzhong were infected by CMV. Meanwhile,the concentration of CMV-Li in ornamental lily leave was higher than that in flowers and bulbs.
