摘要
紫杉醇的生物合成过程已基本阐明,其合成以牛儿基牛儿基二磷酸为前体约需20步酶促反应,近一半的相关酶基因已得到克隆与表达.综述了紫杉醇生物合成及生物合成途径中9个酶基因的克隆和表达方面的研究进展,并指出随着紫杉醇生物合成分子生物学研究的不断深入,利用分子生物技术大规模生产紫杉醇将为期不远.
The path of Taxol biosynthesis from geranylgeranyl diphosphate consists of about 20 enzymatic steps, which has been elucidated approximately. Recent advances on Taxol biosynthesis and the cloning and expression of the 9 genes encoding taxadiene synthase, cytochrome P450 taxdien 5 alpha-hydroxylase, cytochrome P450 taxane 10 beta-hydroxylase, cytochrome P450 taxane 13 alpha-hydroxylase, taxadien-5 alpha-ol-O-acetyl transferase,taxane 2 alpha-O-benzoyltransferase, 10-deacetylbaccatin-Ⅲ-10-O- acetyl transferase, 3-amino-3-phenylpropanoyltransferase and 3'-N-debenzoyl-2'-deoxytaxol N-benzoyltransferase were summarized respectively. The production of Taxol on large scale by biotechnology will become true recently with the development of studies on Taxol biosynthesis in molecular biology.
