牛磺酸减轻兴奋性氨基酸对培养新生大鼠小脑皮层神经元的损害 被引量:8

摘要 取新生大鼠小脑皮层进行体外神经细胞培养,用兴奋性氨基酸建立离体的神经元损害模型,观察了不同浓度牛磺酸对兴奋性氨基酸神经毒性的影响。结果表明:500μmol/L谷氨酸或使君子酸均能造成培养神经元大量死亡,培养液中乳酸脱氢酶含量同步明显增高,在培养液中加入牛磺酸能明显降低神经细胞死亡率,其中以2.0mmol/L浓度效果最为明显。揭示牛磺酸可拮抗兴奋性氨基酸神经毒性。 The effects of taurine on the excitatory amino acid-induced toxicity to cultured neonatal rat cerebellar cortical neurons were observed;The death of the neurons was determined with 0.4%trypan staining and the overall cell injury assessed with the measurement of lactate dehydrogenase(LDH)activity.The findings were as follows:(1)The cultures were treated with 500μmol/L of glutamine and quisquallate in Tris buffer for 5 min and the death rate of the neurons in 24 h was 41.8%±3.5%,47.1%±3.5%respectively.(2)1.0,2.0,4.0 and 6.0 mmol/L of taurine exerted Obvious protection to the cultures and the maximal protection was found when 2.0 mmol/L of taurine was used since the death rate of neurons was decreased to 25.1%±1.9%(Glu group)and 30.6%±3.0%(QA group)respectively.Our findings suggest that taurine can antagonize the neurotoxicity of excitatory amino acids.
